Genetic immunization approaches are particularly suited to a rapid pandemic response. While no genetic immunization vaccine has ever been licensed by the FDA for human use, these platforms hold great promise. Genetic immunization candidates can be seen as mimics of natural infection or of traditional live-attenuated vaccines, like the measles-mumps-rubella vaccine.
We reviewed the literature on host genetic factors related to coronaviruses. We identified 1,832 articles of potential relevance. 75 involved human host genetic factors, of which 35 involved analysis of specific genes or loci; aside from one meta-analysis, all were candidate-driven studies, typically investigating small numbers of research subjects and loci.
109 articles were included. The risks of mortality or severe diseases were higher in patients with increasing age, male gender, dyspnea, diabetes, hypertension, congestive heart failure, hilar lymphadenopathy, bilateral lung involvement and reticular pattern.
